Antioxidant Activity Test of N-Hexane, Ethyl Acetate, and Aqueous Fractions of Santang Mandarin Orange Peel (Citrus reticulata) Using the DPPH (1,1-Diphenyl-2-Picrylhydrazyl) Method

Abstract

This study aims to determine the antioxidant activity of the n-hexane fraction, ethyl acetate fraction, and water fraction from santang orange peel using the DPPH (1,1-Diphenyl-2-Picryl Hydrazyl) method and to determine the IC50 (Inhibitory Concentration 50) value. The Santang orange peel was extracted using the maceration method with ethanol as the solvent, followed by a multi-step fractionation process using solvents with different levels of polarity, namely the n-hexane fraction, the ethyl acetate fraction, and the water fraction. Antioxidant activity assays against DPPH radicals were conducted on the n-hexane fraction, ethyl acetate fraction, and water fraction to determine the comparative inhibitory effects of the antioxidants. The results of the study showed that the ethyl acetate fraction and the water fraction generally had higher antioxidant activity compared to the n-hexane fraction due to the presence of more polar phenolic and flavonoid compounds. The IC50 values obtained represent the concentration required to scavenge 50% of DPPH free radicals; the lower the IC50 value, the higher the antioxidant activity.
